The engineer and the authorities will not allow the aircraft to fly since emergencies might happen and an aircraft slide raft could be necessary for that.

As Aircraft operation is considered yes it can fly as slide raft is used in emergency evacuation procedure and according to MEL considering the safety of passenger civil aviation will not allow you to fly unless slide raft is attached to the aircraft.

If the Rafts are required as part of the MEL. most probably the aircraft will not fly with paid customers

Well, MEL will not allow it to fly if it is full passengers with crew, each escape raft slide according to the FAA certification will carry between 20 to 25 persons or some times as 40 persons as it depends on where it is attached or located, if it is on the doors or in the over heads racks between the escape passengers doors, the slides act as both a slide and a raft boats, SO the aircraft can fly all the times with one or two or three etc slide escape raft is an operative or missing, as far as the number of the passengers and crew been calculated as it is less than the the slides and rafts capacity, as well as the passenger seating locations as regarded to the operative location to the slides and rafts, in case of emergency landing on the ground and sea. life vest under the seat I do think is more safety emergency important for the passengers and the panic situation which is far from to be controlled till now as it is the most reason for casualties during emergencies during commercial flights, because it is a reaction of fear for life and selfishness most of the time, but we have to be ready for it so why we most train and train to get used to it before it does happen in real so we be ready to deal with it calmly, still landing safe over the water on the belly of the aircraft with those two engines hocked to the under wings is more difficult than manually or automatically inflate the slide rafts. wish you all a safe and quite journey NO PANIC PLEASE FROM THE CREW IN THE FIRST PLACE AND PASSENGERS SECONDS AS WELL AS THE CARGO IN LAST AND THIRD.

The MEL / MMEL is the Specific reference Document for such cases. As Long as it comes to permission, usually it is allowed but with penalty.

Most transport category aircraft equipped with inflatable slides/rafts have MEL relief to operate with one or more slides inoperative. This normally requires limiting the number of passengers on board until all slides are serviceable. Commercially this can be troublesome as the available seats for each flight that that specific aircraft is to operate must be restricted in the reservations system and cannot be allow to board passengers above that limit. Logistically and financially it is better to replace the inoperative slides immediately.

Technically, it can fly without a slide raft, but they wont. The reason is its part of the safety requirements and thus it is necessary to have one installed and functioning before any flight. Hope the answer is helpful.
