An effective method I have used to enforce rules and regulations is by giving the person a warning first and politely asking them to stop. If they do not stop, then they are written up or asked to leave depending what is going on. I have then told management about it if it needs further disciplinary actions, but usually once you ask the person to stop, they do so.

I do believe that the effective method for any manager to enforce rules and regulations is to be always " An Ideal Example" for his staff.
An effective method of applying rules and regulations that I have used in project management is the formulation of quality and safety manual at start of project which is finalized with due approval of client. It includes synopsis of the project, organizational structure, quality policy of company, quality procedures and certifications, quality control formats, responsibility and accountability statements, design review, design certification process, vendor review, testing methods, calibration of test equipment, training of staff, document control, record keeping, use of scientific methods and statistical techniques, warranty claim and insurances procedures, safety near miss report format, safety weekly report formats,etc all specific to the current project. These rules and regulations are then applied stringently

In fact there are some set of rules and regulations to organize the work at the offices and work sites and all employees and workers have to respect them to achieve their work efficiently without incidents and problems. The Effective Method to enforce these rules and regulations is in brief in the following points :
1- Let yourself first more committed in front of all to apply these rules and regulations during daily work in the offices and work site
2- Organization meetings and presentations to explain benefits of respect and apply these rules and regulations
3- Presentation some proposals to develop these rules and regulations to be fit with the work environment
4- Set punishments system to any one break these rules and regulation with incentives to anyone more committed in these rules and regulations

I would approach the individual, explain what was going on, and tell them the consequences. 3.Whistle; explaining the rules. 4.I would hold people accountable, and make them serve the consequence no matter who they were. 5.An effective method I used to enforce the rules is looking at everything as positive as I can, you don't want to be the one who right from the start is very negative and bossy. Don't seem like a hardass at first. If someone keeps disrespecting me and the rules then it is time to be a bit harder and make sure everyone understands the rules and consequences.
Other than this there are measures like penalty and punishment , counselling , rewards , motivations etc. can be used .
I would say that the answer depends on the type of regulation or rule we want to enforce. If it is a Health and Security rule (that can put to danger the integrity or even the life of the employees), I believe the only way to make sure it is respected it's to set penalties for those who are breaking it. In case the rule is referring to the company image (behaviour vs customers or suppliers, dress code, etc), more effective would be to set positive examples (such as giving a prize for "the most dedicated employee of the week/month", regardless of the result of his/her work, but only for respecting that specific rule)
