according to the expected size of the fragment that we will obtain and the total size of the organism's genome

it' ranges from100bp to1 kbp

what range you are expecting will  depends on  your sample.  I guess min50bp to max10kb range of markers available.

It really depends on the expected size of your fragment. If you were unsure, maybe run more than one gel. There are ladders of different sizes, e.g.- bp,0.5- kb, etc.

 

Also, some ladders have bands of different masses that are reflected by the intensity of the bands. This enables one to estimate how many grams are present in each band present on the gel.

it depends on the size of the DNA fragment that you will obtain. normally you have to know the fragment size of DNA that you will obtain
