We would require a minimal of following things as for Manual testing is considered

1. Domain knowledge

2. Testing Processes

3. Module Ownership (Applies for everyone)

4. Maximum Coverage during testing

5. Commitment to work (say Attitude)

6. Knowledge on Defect Life Cycle

7. Proper reporting of issues

8. Various modes of finding defects (Say Logs, Recent fix, Impact analysis ....)
