Is there any chance to Claim cost when the variations exceeds 20% of contract value if it is so how?

Yes & No. Yes if the variations were approved and deliverately explains to owner/management.

No- if the contractor or the designer (consultant-MEP as ex:) made a mistake on the design or costing the projects, considering that it was stipulated on the contract/design specs.
