I think the main difference between leader and employee is that leader has the ability to lead people to achieve a specific goal; but the employee has only a particular role in the team

I think the question should be "what is the main difference between leader and follower"

 

And the answer to this question is "Leader sets the target (or vision) and shows and ispires others how to achieve this target. Follower just follows and does his part"

I support my colleague Elie's answer and i will add , that leader even he is not a manager always has a feeling of responsibility toward others colleagues and company, creativity his manner to act between his colleagues , and he hates the sameness always need something new to work in , he is the person who stay after working hours while his colleagues leaving before hours.

A Leader as the meaning of the word is the one who leads by example. Employee/follower follows them.

A Leader initiates the plan of action and employees/followers brings it into action.

A Leader takes risk and brings in innovation, employees/followers rely on leader's decision

Once upon a time today's leader was an employee! Otherwise the leader is the son / daughter of father who was leader and as a heir he/she become a leader. The former was out of efforts but the latter was out of heir status.

On role basis, Leader set the Plans and through his guidance the employees or his followers execute the plan to plan to achieve the common desired goal or vision, "As Steam to a Steam Engine"

The main differences between a leader and employee are the inspiration and the enthusiasm and vision.

The leader is the main source of the inspiration and the enthusiasm for employee, the leader can give both of them to his employee to take the mostly of the employee's effort to achieve the main goals.

The leader has the science and the power to have the right vision of the future of the project so he chose the suitable plan and persons to achive goals in a short way.

Remember the difference  between them in the Holy Qur'an

وَقَالَ لَهُمْ نَبِيُّهُمْ إِنَّ اللَّهَ قَدْ بَعَثَ لَكُمْ طَالُوتَ مَلِكًا ۚ قَالُوا أَنَّىٰ يَكُونُ لَهُ الْمُلْكُ عَلَيْنَا وَنَحْنُ أَحَقُّ بِالْمُلْكِ مِنْهُ وَلَمْ يُؤْتَ سَعَةً مِّنَ الْمَالِ ۚ قَالَ إِنَّ اللَّهَ اصْطَفَاهُ عَلَيْكُمْ وَزَادَهُ بَسْطَةً فِي الْعِلْمِ وَالْجِسْمِ ۖ وَاللَّهُ يُؤْتِي مُلْكَهُ مَن يَشَاءُ ۚ وَاللَّهُ وَاسِعٌ عَلِيمٌ ( ) Al-Baqarah ( The Cow ) - Ayaa

And their prophet said to them, "Indeed, Allah has sent to you Saul as a king." They said, "How can he have kingship over us while we are more worthy of kingship than him and he has not been given any measure of wealth?" He said, "Indeed, Allah has chosen him over you and has increased him abundantly in knowledge and stature. And Allah gives His sovereignty to whom He wills. And Allah is all-Encompassing [in favor] and Knowing

The main difference between a leader and employee is that a Leader should multitask; Act as an employee and behave like a father (Manager), supportive, oversight the way forward, has a Clear Vision and Lead all along to the targeted goal.
