As secretary or assistant I show my initiative to work being always attentive upon what is happening in the office and being always ready to run new tasks.
Companies are always working with deadlines and are, generally, full of work. Thus, my boss needs me to run tasks with agility, but without losing accuracy, as far as he needs me to take some decisions by myself, with moderate autonomy, in order to keep the flow of work.
People who do the things the way they have always been done will in the best case get the same results all over again.
It’s not a secret that even for staying in the same place you have to run faster and faster. Thereby, for standing out you need to be creative. You should constantly search for new solutions and more effective approaches. Ideas are the most expensive matters nowadays and so far the best contribution you can offer to your organization.

just work, work and work
You need to go above and beyond the tasks assigned to you so that you can enhance your manager's work, be self-promotional, consider every opportunity, speak up and share your ideas, and always be prepared.
1. Check if there is anything in the company that needs your initiative.
2. Check if you can make any initiative in the things the company requires.
3. Choose the right time and place and submit your initiative.

In my opinion, one should offer a helping hand when work is piled up. Especially EA as a rule help almost everyone in the office. Usually I just ask what can I help with. And senior staff always have something you can help with. If the product your company is offering is competitive enough you will not have troubles in selling it and on your own initiative to help marketing/sales department to connect with the potential clients in order to sell the product and benefit to your company goals. Whatever you find you can handle with in the office just show your readiness to deal with the stuff and most probably everyone in the office will just happy with your initiative. Just be careful to inform senior staff if you are about to take some more serious initiative in case not mess the things up. :)
