This is really interesting quesiton and many of the job seekers come across such situation. Well first of all my principle is Never Give Up! I will sure apply though I don't have one of the demanded qualifications in the hope that my gained experience and some plus points may be much impressive than the demanded qualification which I do not have. We learn by experiences rather than qualifications.

Its depend on how effectively the eligibility criterias of one suits to the offer placed. Literally speaking, one who does not posess the very qualification of what is being demanded, cannot look into seeking an opportunity by any way.
