Medical diagnostics is a collective field, it has it all from symptomatology to labwork, diagnostic radiology and a whole lot other things to do.
the specific methods are done according to the presenting case, small example to explain if you suspect COPD, from symptoms labwork and xrays, you do a spirometry eventually.
Another two contradicting way, once more depending on the disease, the "see if he gets better, see if he gets worse" way; is diagnostic therapy (as neostygmine in mysthenia gravis patients) and challenges test (basically poking the disease with a stick to confirm it is what we suspect, simply as scratching a patient with urticaria)

Diagnosis based, major complaints, then developed or directed by anamnese, general physical examination. Supportive Investigation such as Laboratory, Radiological, CT Scan, MRI checks are performed, when necessary, appropriately, quickly and quickly.
