We have400 PC's ,45 Servers, Three Forests, ERP System, HR, SharePoint, Automation for industrial and production lines. things are managed and designed in house, we have MIS section and Infrastructure and security section as well as automation of business processes. Is there any standard way of defining the number of employees required to manage IT department there.

Wow, this company has a very good MIS. An analysis is must to determine the exact requirement. To ensure somooth operation a good number of team members for differnt responsibilities are required Lead by a vertern with leadership role. But a good guess 15-20 personnel's are required.
The first and the foremost objective is to get the Job Roles required (This can be done with the help of the IT Head). Then we need to write down the job despcriptions for the aforesaid Job roles.
You need to ensure that the Head of IT is in sync with the Job Descriptions and approves them.
Once these are ready you may give the man power requirement document to the Head of IT and ask him about the Head count requirement, as he is the best one to do so.
The role of HR here is to ensure that there is no over hiring for any role which can be easily done by ensuring that the JD's are precise.
