Simply use Ditributed file system( DFS) in AD for multiple drives or targets. You can permanently set this from the Active Directory User profile settings as a Home Folder.

Mapping network drives and and environment variables can be set using Group Policy preferences in GPOs. which is available in windows server 2008 and above.

certain registry values can also be set using the same.

Nowadays it very easy and simple step to map an network drive.

There is an option in newly relaesed version from 2008 just click map network drive and set path to map that specific and will be mapped like harddisk in your my computer page.

By using GPO....make a GPO in group policy in your server...if you are using virtual environment even then you can add a new GPO with a unc path defined.

Use Group Policy for this and target DFS namespace. This will not break the drive mappings or UNC paths.
